Technological and experimental progress make now possible to measure and perturb systems in various subfields of biology, including neuroscience, genomics, immunology. Exploiting these data, sometimes partial and noisy, and using them to understand and manipulate those systems are fundamental objectives in biology, as well as in bioengineering.  In this talk, I will show some of the works we have done, mostly in the context of protein and RNA modelling and design. I will present the unsupervised learning methods, based on Boltzmann machines, we have developed, and how to make them            efficient and interpretable. Last of all, experimental validations of those approaches and perspectives will be presented.
